Previously piLLai lOkAchAryar explained the main reasons for fully giving up attachment towards worldly pleasures and accepting bhagavath vishayam; in this context, subsequently, he is identifying the main reason for fully giving up other means.

prApakAnthara parithyAgaththukkum agyAna aSakthigaLanRu, svarUpa virOdhamE pradhAna hEthu.
Simple Explanation
Ignorance and inability are not the main reasons to fully give up other means, but being contradictory to one’s true nature is.

upAyams (means) other than prapaththi (surrender).
agyAna aSakthigaL anRu
The main reasons for abandoning these other upAyams are not a) lack of knowledge about them and b) lack of ability to perform them.
svarUpa virOdhamE pradhAna hEthu
The main reason is that those upAyams will not match the true nature of the self which is totally dependent on bhagavAn.
With this, it is implied that ignorance and inability which were identified in sUthram 43 “agyAnaththAlE prapannar” (those who surrender due to ignorance), are not the primary reasons. If one abandons other upAyams due to ignorance and inability, it will imply that when one has knowledge and ability, one can pursue those upAyams. Only when they are abandoned due to being contradictory to one’s true nature, will they never return.
